水稻叶蝉抗性基因回交转育和CAPS标记辅助选择   总被引:10,自引:1,他引:10  
 以综合性状好但对黑尾叶蝉 (NephotettixcincticepsUhler)敏感的品种台中 6 5作为轮回亲本 ,与抗性品种DV85连续回交 ,得到回交高代BC6F2 群体 ,进行抗叶蝉性状的回交转育。将抗黑尾叶蝉基因Grh2两侧的RFLP标记C189和G14 6 5成功地转换为在亲本间具有多态的CAPS标记。在进行表型选择的同时 ,利用CAPS标记对BC6F2 进行了标记辅助选择 ,分析了CAPS标记与Grh2间的遗传距离和标记辅助选择的效果。所选出的个体具有台中 6 5的遗传背景且携带纯合Grh2基因 ,可作为聚合抗叶蝉基因培育新品种的重要中间材料。  相似文献

The firmness of whole beans and of separated embryos and testas from 20 soybean cultivars grown in the same field conditions was estimated after 40 min cooking with AACC International Approved Method 56‐36.01. There were considerable variations in the firmness of whole beans, embryos, and testas among cultivars. Testas were much firmer than embryos because of their leathery texture and resistance at slots in the shear cell during compression. For soft cultivars, the firmness after 10 min cooking was also measured, and an apparent first‐order reaction kinetics model was applied to estimate the initial firmness at 0 min cooking and the softening rate constant from firmness data after 10 and 40 min cooking. The estimated initial firmness at 0 min cooking and softening rate constant of embryos and testas varied widely among seven cultivars, and these two parameters were considered to be controlled by different genetic mechanisms, because no correlation was found between the parameters. The softening rate constants of testas were much smaller than those of embryos, and the relative firmness of testas to embryos increased during cooking. The firmness of whole beans and embryos after 10 min cooking would be a useful characteristic for approximating their firmness after 40 min cooking.  相似文献

The lentinan contents in the Lentinus edodes fruit body during storage were examined by ELISA method using anti-lentinan antibodies. The lentinan content (12.8 mg.g(-)(1) dw) before storage decreased to 3.7 mg.g(-)(1) dw over 7 days at 20 degrees C. However, it only slightly decreased at 1 degrees C and only decreased to 9.3 mg. g(-)(1) dw at 5 degrees C. Glucanase activity, which seems to be associated with lentinan degradation, increased more during storage of L. edodes at 20 degrees C than it did at lower temperatures. In addition, only glucose was detected as a degraded product from lentinan by the glucanase. This suggested that this enzyme would fit the profile of an exo-type glucanase. Also, polyphenol oxidase activity, known as an index of freshness reduction in the mushroom, increased approximately 2.7-fold (to 61.5 units.mg(-)(1)) over 7 days during storage at 20 degrees C. However, its activity changed little during storage at lower temperatures. These results indicate that the reduction during storage of the quality of L. edodes as a functional food is accompanied by the decrease of lentinan, and by browning, and that exo-glucanase plays an important role in the decrease of lentinan content.  相似文献

Onions (Allium cepa L.) are produced in many countries and are one of the most popular vegetables in the world, thus leading to an enormous amount of international trade. It is currently important that a scientific technique be developed for determining geographic origin as a means to detect fraudulent labeling. We have therefore developed a technique based on mineral analysis and linear discriminant analysis (LDA). The onion samples used in this study were from Hokkaido, Hyogo, and Saga, which are the primary onion-growing areas in Japan, and those from countries that export onions to Japan (China, the United States, New Zealand, Thailand, Australia, and Chile). Of 309 samples, 108 were from Hokkaido, 52 were from Saga, 77 were from Hyogo, and 72 were from abroad. Fourteen elements (Na, Mg, P, Mn, Co, Ni, Cu, Zn, Rb, Sr, Mo, Cd, Cs, and Ba) in the samples were determined by frame atomic adsorption spectrometry, inductively coupled plasma optical emission spectrometry, and inductively coupled plasma mass spectrometry. The models established by LDA were used to discriminate the geographic origin between Hokkaido and abroad, Hyogo and abroad, and Saga and abroad. Ten-fold cross-validations were conducted using these models. The discrimination accuracies obtained by cross-validation between Hokkaido and abroad were 100 and 86%, respectively. Those between Hyogo and abroad were 100 and 90%, respectively. Those between Saga and abroad were 98 and 90%, respectively. In addition, it was demonstrated that the fingerprint of an element pattern from a specific production area, which a crop receives, did not easily change by the variations of fertilization, crop year, variety, soil type, and production year if appropriate elements were chosen.  相似文献

Starch and gluten were isolated from 10 wheat cultivars or lines with varied amylose content. The rheological properties of 30% wheat flour gel, starch gel, and the gel of isolated gluten mixed with common starch were determined in dynamic mechanical testing under shear deformation, creep‐recovery, and compression tests under uniaxial compression. Variation of wheat samples measured as storage shear modulus (G′), loss shear modulus (G″), and loss tangent (tan δ = G″/G′) was similar between flour and starch gels and correlated significantly between flour and starch gel. The proportion of acetic acid soluble glutenin exhibited a significant relationship with tan δ of gluten‐starch mixture gel. The small difference in amylose content strongly affected the rheological parameters of flour gels in creep‐recovery measurement. Wheat flour gel with lower amylose content showed higher creep and recovery compliance that corresponded to the trend in starch gel. Compressive force of flour gel at 50 and 95% strain correlated significantly with that of starch gel. Gel mixed with the isolated gluten from waxy wheat lines appeared to have a weaker gel structure in dynamic viscoelasticity, creep‐recovery, and compression tests. Starch properties of were primarily responsible for rheological changes in wheat flour gel.  相似文献

Wheat lines with reduced amylose content were recently produced by single and double mutation from a low‐amylose line, Kanto 107. They are appropriate for clarifying the influence of amylose content on starch gel properties because of their similar genetic background. When measured using the concanavalin A method (ConA), the total amylose content of isolated starches from Kanto 107 and three mutants (K107Afpp4, Tanikei A6599‐4, K107Wx2) was 24.8, 18.5, 7.1, and 1.7%, respectively. Results of differential scanning calorimetry (DSC) showed that the difference in amylose content strongly affected gelatinization conclusion temperature and enthalpy. We prepared 30 and 40% starch gels and measured their dynamic shear viscoelasticity using a rheometer with parallel plate geometry. Compressive and creep‐recovery tests were conducted under uniaxial compression. The storage shear modulus correlated highly with the amylose content of starch in 30 and 40% starch gels. The creep‐recovery test showed a clear distinction in creep curves among starch samples. When the compressive force required for 50, 80, and 95% strains was compared, starch gels with lower amylose content showed lower compressive force at 50% strain. Waxy starch gel (K107Wx2) showed higher compressive force at strain >80% than other samples due to its sticky property.  相似文献

Pernambuco (Guilandina echinata Spreng. syn Caesalpinia echinata Lam.), which has been used for a material of violin bows, was compared with substitutable wood species from the viewpoint of processing properties and durability. The properties required for bow material are discussed. The shearing strength of pernambuco was higher than that of the other wood species at the same specific gravity. High shearing strength seems to be effective for preventing damage to the head (top part) of the bow. The presence of extractives affects the thermal softening of wood material, but ease of handling and permanent retention of form are not particularly superior for pernambuco, although it does have a high extractives content. A peculiarly low loss tangent (tan S) of pernambuco can probably be attributed to the large amount of extractives, rather than the mean microfibril angle.Part of this report was presented at the 46th Annual Meeting of the Japan Wood Research Society in Kumamoto, April 1996.  相似文献

Cassava is a major staple, bio‐energy and industrial crop in many parts of the developing world. In Southeast Asia, cassava is grown on >4 million ha by nearly 8 million (small‐scale) farming households, under (climatic, biophysical) conditions that often prove unsuitable for many other crops. While SE Asian cassava has been virtually free of phytosanitary constraints for most of its history, a complex of invasive arthropod pests and plant diseases has recently come to affect local crops. We describe results from a region‐wide monitoring effort in the 2014 dry season, covering 429 fields across five countries. We present geographic distribution and field‐level incidence of the most prominent pest and disease invaders, introduce readily‐available management options and research needs. Monitoring work reveals that several exotic mealybug and (red) mite species have effectively colonised SE Asia's main cassava‐growing areas, occurring in respectively 70% and 54% of fields, at average field‐level incidence of 27 ± 2% and 16 ± 2%. Cassava witches broom (CWB), a systemic phytoplasma disease, was reported from 64% of plots, at incidence levels of 32 ± 2%. Although all main pests and diseases are non‐natives, we hypothesise that accelerating intensification of cropping systems, increased climate change and variability, and deficient crop husbandry are aggravating both organism activity and crop susceptibility. Future efforts need to consolidate local capacity to tackle current (and future) pest invaders, boost detection capacity, devise locally‐appropriate integrated pest management (IPM) tactics, and transfer key concepts and technologies to SE Asia's cassava growers. Urgent action is needed to mobilise regional as well as international scientific support, to effectively tackle this phytosanitary emergency and thus safeguard the sustainability and profitability of one of Asia's key agricultural commodities. © 2016 Society of Chemical Industry  相似文献
